Fruity Fruity Fruit Juice
Introduction:
If you have bought fruit juice from a drinks stall or fruit stall before, then you would most definitely have seen this fairly large and noisy blending machine that turn your fruits into fruit juice. This conventional method is both tiring and troublesome. Not only do we have to press the fruits which accumulate in the machine when the fruits are blended. In this experiment, we will be using an easier way to get your fruit juice. We will not be using machine, but instead enzyme solution.

Aim: To use different solutions to extract fruit juice and find out the most effective method.

Material:
- Four 100cm3 Beakers (Labeled P, A, C, D)
- Filter Funnel
- 4 Filter Papers
- 4 Measuring Cylinder
- 1 spatula of Pectinase Enzyme (P)
- 1 spatula of Amylase Enzyme (A)
- 1 spatula of Cellulase Enzyme (C)
- 5 cm3 Distilled Water (D)
- 4 Droppers
- 4 Oranges
- Blender
- Stop Watch
- Glass Rod

Procedure:
1. Blend an orange using the blender
2. Pour the blended orange juice into one of the beakers. Repeat this for the other 3 oranges.
3. Label the beakers (P, A, C, D)
4. Add 1 spatula of each enzyme into their corresponding beakers (P, A, C) and 5cm3 of distilled water (without enzyme) into the beaker labeled D.
5. Stir the contents in each beaker with a glass rod and leave it to stand for a few minutes.
6. Fold a piece of filter paper and put it into a filter funnel. Place the filter funnel on a measuring cylinder.
7. Filter the contents of one of the beakers by pouring them into the funnel.
8. Record the volume of juice collected in the measuring cylinder.
9. Repeat steps 6 to 8 for the other 3 beakers.

In Beaker P, we can find the largest volume of orange juice as Pectinase Enzyme break down the cell walls of oranges’ cells, releasing more orange juice inside the cells. In the factory, they used enzymes to produce more fruit juice.

16th June 2008 (1st Day )♥♥♥

Today I went for the 2 Days Web Designing Workshop. It is held at a place near Tanjong Pagar MRT station. So, it is quite far from my home. We learn how to design our own website with Adobe Photoshop and Dreamweaver. But, today, we only learn on the Adobe Photoshop. This course provides you with the concepts and skills to use Adobe Photoshop effectively. You will get hands-on experience working with basic and advanced image editing techniques. You will learn colour painting, photo retoching and image eniting. Is also strongly oriented toward design details and functions;it also explores the realities that apply to the use of Photoshop.Colour correction and adjustment are stressed in various projects as well issues of output for printing.
Today, when I reached there, I reached at exactly 9am lor. Wah..I so punctual ah... Just Joking. Anyway, when I reached there, I was shocked thaqt I found some of my friends from the Operation:SMART. :) Anyway, I sat beside Li Yang. Wah... What bad luck. just after lunch, the harddisk don't know why fell on the ground and the computer then restart on cannot go back into the desktop then it said something was missing??? Something were missing just when the harddisk fell on the floor??? IMPOSSIBLE! Anyway, then before the 'technicial' came and 'fix;, i can still move some of the things. But, after the 'technicial' 'fix' it, the whole computer could not move!!! Hiaxx... But, I have fun today. Ok lah. bb tomoorow still got lesson/.♥♥♥

17th June 2008 (2nd Day )♥♥♥

Hi there! As I had told you in my last post, I went to the Web Designing Workshop. Anyway, today we learn about the Dreamweaver. It was fun today. But, today, I didn't reached the Singapore Technologies Building at exactly 9am today. Dreamweaver is a leading Web page layout and site management program. Designers and developers both use this software to efficiently create and manage beautiful static and dynamics web sites. We could try the different way to edit the HTML which is Hyper Text Markup Language. By the way, today I could use a computer that had no problem. Phew... In this time, we use the software 'Dreamweaver 8'. There are in total three type of view for us to see. They are 'Code View', 'Split View' and 'Design View'. In the 'Code View', we can apply HTML tags to design their website. By using HTML tags, we can edit fonts, tables, alignments and etc. In the 'Split View', we can get to view both code and design view together while designing their website. The advanatge of this view is that we can either edit their website by adding HTML in the 'Code View' or directly edit on the 'Design View'. In the 'Design View', we can directly typed text or insert pictures inside the view. the view that the users see over here will be the same as the end result when users publish it on to the Internet. There are also many things for us to learn though. But, anyway, I like today's lesson that yesterday's. The lesson today is so much fun and we could get to edit the HTML and much more. :) It was really fun. Okay, that's all. Post next time. :)♥♥♥

Opening of the Fo Guang Shan

佛光山
(8th June 2008)

佛光山
Hi there! It's me again! On 8th June 2008, I went to the Opening of the Fo Guang Shan. It was located at Punggol so I just need to take around 5 minutes in the bus 82. When I reached there, I had a big shock. There are a lot of people there. As it is still early, the people there did not allow us to go in into the temple. But, from the outside, we could see that the temple was really big and nice. It is painted with white. Then, I went to the opposite side of the road. There is another event there from Fo Guan Shan. There is free vegetarian food there too. But, from the packaging, it looked more like Japanese food, only that it is vegetarian... :P Anyway, there is also pudding. Yum Yum! Anyway, do you want to know what is in the vegetarian food I had gotten? Do not worry as I will tell you. :) Anyway, there is a pile of yummy noodles, then vegetarian-made meat only that one of them is fried and another is with tomato sauce. Then, there is two sushi. Yummy! In the whole packaging, I like the sushi the most. I think you will like the sushi too if you had gotten a chance to eat it. Then, there are a lot of steam peas. Of course, to let the meal be balanced, there are fruits... There are a piece of Guava, a grape and dried papaya. Then, I saw something amazing. It is ginger that is PINK!!! But, of course, I didn't eat it. I don't like ginger though, as you know. :) Anyway, there are also some free souvenirs. Woah! The souvenir was so nice! It is a crystal ball with keychain. And then, in the crystal ball, there is the Buddha and the word'佛光山' on it. And it was like made of tiny bubbles. I like it very much. It is put in a red box. Anyway, there is also a something like a concert and some people are singing. Finally, it was 1.30p.m., and we all can go into the temple. Do you know how many levels are there in Fo Guang Shan? Let me tell you! There are a total of 4 levels!!! I started ‘exploring’ from the 4th level. There are restaurant there for the people there. On the 3rd floor, there is a place for the people to eat and the 2nd floor is the library. When my father took a book of the history of the Fo Guang Shan and with the three souvenirs and took the picture, the people there was like kept looking at him. Then, an auntie (about my grandmother’s age) came to ask my father where he had gotten the souvenirs. As my father was busying taking photos, he told the place. Then, the auntie rushed off without listening all what my father had said. If she had listened, she would not need to waste her effort to go there. The sentence she didn’t listen is that the souvenirs all had been given. Okay, that’s all bye!

Hi there everyone! It's me again. Hiaxx… Today I still need to go to school. Actually, as I had told you all, that today I had Life Science Enrichment programme on DNA. Actually it's quite fun. It is not as boring as I had thought. We got to do 2 hands-on activities .Yeah! Okay, I’ll tell you what we had done in today’s activity.

Even though this Life Science Programme was started at 0900 hours, we must reach there at 0850 hours. After that, I went up to the hall together. We then sat down according to our class. Then, the teacher said that each class needs to sit in two rows. Then, the teacher started the lecture on DNA. But, before that, the chair person had to give out a pile of worksheet and mark the attendance. From the lecture I had learned a few things and I will tell you some information on the DNA. DNA stands for Deoxyribonucleic Acid. The basic structure of DNA is a sugar phosphate backbone and two strands form a helical structure (A double helix). The four nitrogenous strands bases in DNA are Adenine (A), Thymine (T), Cytosine (C) and Guanine (G). Adenine will always pairs with Thymine and Cytosine will always pairs with Guanine. They are held together by hydrogen bonding. DNA is found at the nucleus of the cell. 46 chromosomes are found in a normal human being. DNA provides instructions for making proteins. DNA determines the features and characteristic of living organisms. After the lecture, different classes went to different lab. We went to the Physic Lab 2, which is at Level 4.

Then, we started to do the first activity which is ‘Making an Edible DNA Model’. For this activity, we need to have 2 Licorice sticks, Coloured gummies – 9 green, 9 pink, 9 yellow, and 9 orange, 9 toothpicks and a plastic container. First, we choose a sequence and write the complementary sequence. The first sequence is T A C G T A T G A and the second sequence is T G G T T T A G A. I chose the second sequence. Then, we assemble one side of our DNA molecule. A piece of licorice will form the backbone and gummies will be the chemical bases. According from Adenine (A) = Yellow gummies, Thymine (T) = Pink gummies, Cytosine (C) = Green gummies and Guanine (G) = Orange gummies. We then place a gummy on the end of the toothpick goes all the way through. Anchor the toothpick into the licorice backbone. Then, we match the chemical base pairs. Place the colored gummy for the matching chemical base on the other end of each toothpick. We complete our DNA model by attaching the other backbone so our model looks like a ladder. We then twist our DNA model carefully so that it looks like a double helix. Actually, Mr. See did come and see us. He told us not to eat the gummies as it is poisonous. -_-||| Actually, the teacher had already told us not to eat as it is put at lab, don't know for how long. :)

We then do the second activity which is the extraction of DNA from strawberries. Actually, we need to do this in a pair. But, I had no one to pair up so I do the activity. Then, I found out that it is not a really bad thing after all. Those people who are doing this activity in a pair had to share the DNA while I got to bring home all the strawberry’s DNA. Yeah! First, we need a strawberry, a Ziploc bag, 15ml of buffer solution, ethanol, filter, beaker, boiling tube, glass rod, and a dropper. We must be careful with Ethanol as it is very flammable and we must keep away from flames and sparks. First, we put the strawberry in a Ziploc bag. Seal the bag while pushing the air out of the bag and use our fingers and hand to crush the fruit for 1-2 minutes. Some of them were not crushing the fruit but hitting the fruit lor. Then, add 15ml of buffer solution to the bag and reseal the bag. Mix the mixture with our fingers for another 1-2 minutes. Then, pour the mixture from the bag through the filter over a beaker. Use a glass rod to gently move the mixture around to allow liquids to pass through the filter into the beaker. Pour the mixture from the beaker to a test tube and collect 2-3ml of the solution. Pour the alcohol until we see a layer of alcohol above the solution. Allow the test tube to stand for 2-3 minutes. We will observe white jelly-like substance floating in the alcohol. The white jelly-like substance is the DNA of the strawberry. Using a dropper, we fill our micro-tube with alcohol. We then collect the DNA using the micro-tube of alcohol and put a string through the micro-tube cap and we could bring it home! Yeah! That’s all for what we had did. Okay, that’s all. Bye! I will post again so please wait for my next post!!!!
